This position may assist with several projects in the lab. In one project, we are trying to understand the molecular mechanism of action of a selfish genetic element that consists of a toxin and its antidote. In another project, we are working on mechanisms of neuromodulatory signaling and release of peptide hormones. POSITION PURPOSE The RSE1 position (also known as an entry level "Research Technician") will typically involve a combination of research activities and lab management activities. The ideal candidate may be relatively new to research with some experience, but is expected to transition into a role of more limited supervision after initial training. The position will typically will have their own independent research projects as well as supporting other projects in the lab. DUTIES AND RESPONSIBILITIES Designing and performing experiments (80%). The duties of the position will be to perform experiments deciphering the molecular mechanism of the peel-1/zeel-1 antidote of C. elegans, as well as providing support to deciphering the function of the Rab2 effector RUND-1 in dense-core vesicle biogenesis. Techniques used will include molecular biology (PCR, cloning, CRISPR/Cas9), protein expression and purification, protein activity assays, fluorescence confocal microscopy, and genetics. Specific tasks will include expressing and purifying proteins such as PEEL-1, ZEEL-1, PMPL-1, RAB-2, RUND-1, and TBC-8; performing PEEL-1 activity assays and binding to PMPL-1 and ZEEL-1; performing RAB-2 GTPase activity assays; performing structure-function analysis of PEEL-1, PMPL-1, and ZEEL-1 through mutagenesis and functional assays in C. elegans and mammlian cells; performing structure function of RUND-1 similarly. Meetings and seminars (10%) This position will involve attending a weekly lab meeting and other relevant meetings and seminars related to the field of the work. General tasks to support the lab (10%) A small portion of the time will be spent on tasks to support the overall function of the lab, including tasks such as ordering, equipment maintenance and repair. MINIMUM REQUIREMENTS Bachelor's Degree in Biochemistry or Biology and one year of relevant laboratory work, OR equivalent combination of education/experience. ADDITIONAL REQUIREMENTS Previous work in a laboratory A practical understanding of basic methods in molecular biology Strong organizational skills Ability to learn new research techniques Excellent oral and written communication skills Ability to work as part of a team in a collaborative environment Demonstrated ability to hold position of responsibility for extended period of time Additional Requirements Experience with molecular biology methods including cloning and PCR Experience with protein expression and purification, including protein gels and Western blotting Experience with protein activity assays such as BCA assays.
